RESUMO

The genus Trichoderma comprises more than 500 valid species and is commonly used in agriculture for the control of plant diseases. In the present study, a Trichoderma species isolated from Scleronema micranthum (Malvaceae) has been extensively characterized and the morphological and phylogenetic data support the proposition of a new fungal species herein named Trichoderma agriamazonicum. This species inhibited the mycelial growth of all the nine phytopathogens tested both by mycoparasitism and by the production of VOCs, with a highlight for the inhibition of Corynespora cassiicola and Colletotrichum spp. The VOCs produced by T. agriamazonicum were able to control Capsicum chinense fruit rot caused by Colletotrichum scovillei and no symptoms were observed after seven days of phytopathogen inoculation. GC-MS revealed the production of mainly 6-amyl-α-pyrone, 1-octen-3-ol and 3-octanone during interaction with C. scovillei in C. chinense fruit. The HLPC-MS/MS analysis allowed us to annotate trikoningin KBII, hypocrenone C, 5-hydroxy-de-O-methyllasiodiplodin and unprecedented 7-mer peptaibols and lipopeptaibols. Comparative genomic analysis of five related Trichoderma species reveals a high number of proteins shared only with T. koningiopsis, mainly the enzymes related to oxidative stress. Regarding the CAZyme composition, T. agriamazonicum is most closely related to T. atroviride. A high protein copy number related to lignin and chitin degradation is observed for all Trichoderma spp. analyzed, while the presence of licheninase GH12 was observed only in T. agriamazonicum. Genome mining analysis identified 33 biosynthetic gene clusters (BGCs) of which 27 are new or uncharacterized, and the main BGCs are related to the production of polyketides. These results demonstrate the potential of this newly described species for agriculture and biotechnology.

O uso de bactérias na promoção do crescimento e no controle biológico de doenças em plantas pode minimizar a contaminação ambiental causada pela aplicação indiscriminada de pesticidas e fertilizantes químicos. Objetivamos avaliar a promoção do crescimento e o controle biológico de Corynespora cassiicola em mudas de tomate mediadas por bactérias benéficas isoladas de solo amazônico não-rizosférico contendo diferentes dosagens de biocarvão, e identificar a quais grupos de bactérias os isolados pertencem. Obtivemos 200 isolados de parcelas experimentais contendo doses de biocarvão de 0, 40, 80 e 120 t ha-1. Destes, 53 foram selecionados por testes de colonização radicular. Com base nos parâmetros de promoção do crescimento, 25 isolados foram selecionados, identificados através de análise molecular e avaliados para produção de ácido indolacético (AIA), solubilização de fosfato e controle biológico. A melhor dose de biocarvão para a formação de colônias foi 40 t ha-1, e um modelo de regressão indicou 34 t ha-1como dose ótima. A produção de AIA foi observada em 18 (75%) isolados e dois (8%) isolados foram capazes de solubilizar fosfato. A eficiência na promoção do crescimento das raízes foi de até 125%, e a porcentagem de proteção das plantas variou de 50,3 a 59,0%. A caracterização molecular indicou que as bactérias utilizadas nesse estudo pertencem aos gêneros Bacillus e Lysinibacillus.(AU)

The habanero chilli pepper, Capsicum chinense is an important crop in the Amazon Basin, mainly grown by small-scale producers. Capsicum chinense plants in an experimental field in the northern Brazilian state of Amazonas were found exhibiting characteristic symptoms of viral infection. Leaf sap from symptomatic plants examined under a transmission electron microscope revealed the presence of elongated flexuous particles and isometric particles. Using molecular assays, the viruses were identified as pepper yellow mosaic virus (PepYMV) and cucumber mosaic virus (CMV). Aphids, identified as Aphis gossypii, were found colonizing the C. chinense plants in the field and may be the vector for both PepYMV and CMV. We report the first occurrence of these viruses infecting C. chinense in the state of Amazonas.


A pimenta-de-cheiro, Capsicum chinense é uma cultura importante na Bacia Amazônica, cultivada principalmente por pequenos produtores. Plantas de C. chinense em um campo experimental localizado no norte do estado brasileiro do Amazonas, foram encontradas apresentando sintomas característicos de infecção viral. Extratos de amostras de folhas sintomáticas examinados ao microscópio eletrônico de transmissão revelaram a presença de partículas alongadas e flexuosas e de partículas isométricas. Análises moleculares permitiram identificar a presença do pepper yellow mosaic virus (PepYMV) e do cucumber mosaic virus (CMV). Pulgões, identificados como Aphis gossypii foram encontrados colonizando pimenteiras-de-cheiro neste campo experimental e podem representar o provável vetor de PepYMV e CMV. Este trabalho relata a primeira ocorrência desses vírus infectando C. chinense no estado do Amazonas.

The Anthropogenic Amazon Dark Earth soil is considered one of the world's most fertile soils. These soils differs from conventional Amazon soils because its higher organic content concentration. Here we describe the metagenome sequencing of microbial communities of two sites of Anthropogenic Amazon Dark Earth soils from Amazon Rainforest, Brazil. The raw sequence data are stored under Short Read Accession number: PRJNA344917.

Black Sigatoka disease caused by Mycosphaerella fijiensis is the most severe banana disease worldwide. The pathogen is in an invasive phase in Brazil and is already present in most States of the country. The potential of 29 isolates of Trichoderma spp. was studied for the control of black Sigatoka disease under field conditions. Four isolates were able to significantly reduce disease severity and were further tested in a second field experiment. Isolate 2.047 showed the best results in both field experiments and was selected for fungicide sensitivity tests and mass production. This isolate was identified as Trichoderma atroviride by sequencing fragments of the ITS region of the rDNA and tef-1α of the RNA polymerase. Trichoderma atroviride was as effective as the fungicide Azoxystrobin, which is recommended for controlling black Sigatoka. This biocontrol agent has potential to control the disease and may be scaled-up for field applications on rice-based solid fermentation.


A Sigatoka-negra causada por Mycosphaerella fijiensis é a doença mais destrutiva da bananeira em termos mundiais. O patógeno está em uma fase invasiva no Brasil e já se encontra distribuído na maior parte dos Estados do país. O potencial de 29 isolados de Trichoderma spp. para o controle da Sigatoka-negra foi estudado sob condições de campo. Quatro isolados foram capazes de reduzir significativamente a severidade da doença e foram selecionados para um segundo experimento de campo. O isolado 2.047 apresentou os melhores resultados e foi utilizado em testes de sensibilidade a fungicidas e produção massal. Esse isolado foi identificado como Trichoderma atroviride por meio do sequenciamento de fragmentos da regiões ITS do rDNA e tef-1α da RNA polymerase. Trichoderma atroviride foi tão efetivo no controle da Sigatoka-negra quanto o fungicida Azoxystrobin, que é recomendado para o controle da doença. O agente de controle biológico tem potencial para o controle da Sigatoka-negra e pode ser produzido em massa em arroz autoclavado para aplicações no campo.

The endophytic niches of plants are a rich source of microbes that can directly and indirectly promote plant protection, growth and development. The diversity of culturable endophytic fungi from stems and branches of Theobroma cacao (cacao) and Theobroma grandiflorum (cupuaçu) trees growing in the Amazon region of Brazil was assessed. The collection of fungal endophytic isolates obtained was applied in field experiments to evaluate their potential as biocontrol agents against Phytophthora palmivora, the causal agent of the black-pod rot disease of cacao, one of the most important pathogens in cocoa-producing regions worldwide. The isolated endophytic fungi from 60 traditional, farmer-planted, healthy cacao and 10 cupuaçu plants were cultured in PDA under conditions inducing sporulation. Isolates were classified based upon the morphological characteristics of their cultures and reproductive structures. Spore suspensions from a total of 103 isolates that could be classified at least up to genus level were tested against P. palmivora in pods attached to cacao trees in the field. Results indicated that ∼70% of isolates showed biocontrol effects to a certain extent, suggesting that culturable endophytic fungal biodiversity in this system is of a mostly mutualistic type of interaction with the host. Eight isolates from genera Trichoderma (reference isolate), Pestalotiopsis, Curvularia, Tolypocladium and Fusarium showed the highest level of activity against the pathogen, and were further characterized. All demonstrated their endophytic nature by colonizing axenic cacao plantlets, and confirmed their biocontrol activity on attached pods trials by showing significant decrease in disease severity in relation to the positive control. None, however, showed detectable growth-promotion effects. Aspects related to endophytic biodiversity and host-pathogen-endophyte interactions in the environment of this study were discussed on the context of developing sustainable strategies for biological control of black-pod rot of cacao.

Visando conhecer a diversidade e a incidência de fungos emboloradores e manchadores de madeira da região Amazônica, foi realizado um levantamento em 12 espécies florestais, estocadas em quatro indústrias madeireiras de Manaus. De cada espécie florestal selecionaram-se cinco toras, das quais retiraram-se amostras de onde procedeu-se isolamento dos fungos. Foram isolados 106 fungos associados às essências florestais, representados por nove gêneros e por dezesseis espécies. Paecilomyces variotii e Lasiodiplodia theobromae foram as espécies fúngicas mais freqüentemente associadas às espécies florestais. Ceiba pentandra e Hura crepitans foram as essências florestais que apresentaram maior diversidade de fungos, com sete espécies cada uma. Hymeneae courbaril foi a que apresentou menor diversidade de fungo, com apenas uma espécie.


In order to know a diversity and incidence of wood mould and wood stain fungi from Amazonian wood, the survey and identification of these kinds of fungi were carried out in twelve wood species from four wood industries of Manaus, Amazonas. Five logs were randomly selected. Wood samples were prepared and transported to the Wood Pathology Laboratory at the Forest Product Research Center of the National Institute for Amazonian Research, where the isolation and identification of the fungi were conducted. One hundred and six fungi associated to the wood species were isolated, represented by nine genera and sixteen fungi species. Paecilomyces variotti and Lasiodiplodia theobromae were the more representative fungi, associated with ten and seven wood species respectively. Ceiba pentandra e Hura crepitans were the wood species with highest diversity of fungi, with seven species each. Hymenae courbaril was the wood that presented the lowest diversity of fungi, with only one fungus.
